Product Description | This product is designed to detect the activity for variety of kinases (see Table 1) and screening the inhibitors of the corresponding kinase. The assay kit is non-radioactive but colorimetric. The principle of the assay is that: The kinase substrates is immobilized onto the 96-well plate. The kinases use ATP to phosphorylate the substrate on the plate. The phosphorylated product is detected by the phosphor-substrate specific antibodies. See figure1 for the principle illustration. |

测试盒原理:
基于ELISA的基本原理。PKC底物被包被在酶标板上,在ATP存在的前提下,PKC激酶将底物磷酸化,洗掉游离的PKC激酶和其他成分。再加入特异性磷酸化抗体(只识别磷酸化底物,而与非磷酸化底物无交叉反应。)检测残留在酶标板上的磷酸化底物。通过酶标二抗放大信号,读出OD值(蛋白激酶活性与OD值的对数成正比)。与对照组相比,得出处理组的蛋白激酶活性。
